Streaking Plates
Purpose:
To recover strains that are frozen for later experiments.
Protocol:
Take strains out of freezer(at -80 degrees C). Gently dig
into the frozen strains using pointed side of streaking instrument.
Streak strains on CM agar petrie dishes (see diagram below). Put petrie
dishes in plastic bag, upsidedown to prevent condensation from dripping
onto the strains. Store in incubator at 37 degrees C for 10 days.
