Making Cultures
Plate to Liquid Cultures
Purpose:
To harvest colonies from a streaked NRC-1 plate to grow Halobacterium in a flask for experiments in the future.
Protocol:
Add 50ml of media(CM) to a 125ml flask. Then find a single colony
from a streaked plate of Halo. Gently use pipette tip
to remove colony. Drop tip in flask, swirl around, and cover the
flask. Incubate at 220 RPM and 37 degrees Celcius. Full culture should
be ready in about 6 days.
Liquid to Liquid Cultures
Purpose:
To grow Halo in a flask for later experiments without using a streaked plate.
Protocol:
Add 50ml of CM to a 250ml flask. Add .5ml of any liquid Halo
culture. Incubate at 220 RPM and 37 degrees C for 48 hours.
