Gas Vesicle Isolation Lab

THE PROCEDURE:

This lab was lead by Megan Meislin to teach the interns what gas vesicles are and why Halobacterium need them. To start the lab the students added 2 mL of Halobacterium to an agar plate. Then they used beads to spread the Halobacterium evenly across the agar plate. After that was done they wrapped the plate with saran wrap to prevent the plate to dry. They incubated the plates for 10 days to allow Halobacterium to really grow.

Using a Pasteur pipette, we recovered the intact vesicles that were floating at the top. We purified the solution by centrifugation until we got pure gas vesicles
