Metals Lab Protocol

1. There should be 25 test tubes for the following metal; Iron Sulfate (FeSO4). Label 25 Falcon tubes as following: Name, Metal, Test Tube (blank, one, two, and three.

2. Pipette 5mL of complete media into all test tubes.

3. Add the respective amount of metal into the four test tubes labeled for that specific metal concentation.

Concentration 0mM 5mM 6.5mM 7.5mM 8.5mM
FeSo4 0 μl 25 μl 32.5 μl 37.5 μl 42.5 μl

4. Add 100 μl of Halobacterium to every test tube of every metal concentration except for the test tubes labled blank for each metal concentration.

I5. Incubate test tubes for 48hrs in 37º C and 220 rev/min (rpm) conditions.

6. Add 1 ml from test tubes removed from the incubator into cuvettes except for blanks, from which you add 1ml into two cuvettes, (the blanks will be used as the reference.)

7. Using the spectrophotometer, measure the cell density of the concentration.

8. Record and plot your data.

9. Clean up your lab space and materials
